Dr. Joerg Dreessen Novartis
Institutes for Biomedical Research, Discovery Technologies / DTI, Basel,
Switzerland was awarded "Best Poster" at the annual meeting of the Society
for Biomolecular screening in Orlando Florida. Dr. Dreessen's Poster "
Rapid Homogenization of Single Step Dilutions, in Miniaturized 1,536 MTP
Liquid Handling" described using a V&P pin tool and an orbital shaker to to
transfer and mix solutions in a 1,536 Microplate. The Poster was also
accompanied by a
movie that we
have placed on the web. Poster Abstract:
Assay miniaturization into 1536MTP, accompanied with low volume handling, is
one important step in pharmaceutical industry on increasing the probability in finding novel
biologically active molecular
entities by continuously increased HTS. This volume range (nano- to
microliter) is physically a challenge (surface tension, diffusion, evaporation, viscosity, factored by
surface/volume ratio).
Unlike other low volume liquid handlers, pin tools provide the opportunity
in low relative dead volume
to transfer compound solution directly from stock to assay. High speed video
visualization of the compound transfer (will be presented) revealed localized inhomogeneous high
compound/DMSO concentrations in the assay volume, which could end up in misleading
results, especially in cellular assays.
A recently launched high frequency shaker prototype has been improved in
collaboration with a leading manufacturer for a) mixing efficiency and b) use in fully automated systems,
resulting in homogeneous distribution of compound/DMSO in a 1536MTP well in fractions of a second.
